Looks like you are using VMware Virtual Disk Development Kit (VDDK) version 6.0. VDDK 6.0 is very buggy and actually VMware Virtual Disk Manager in VDDK 6.0 does not even work. This is why you are getting the " Can't create connection." Error.

Try installing and using VDDK 5.1.4 and 5.5.4 (together) and you should be good to go. The vBackup Administrator's Guide covers the install of VDDK 5.1.4 and 5.5.4 and using them both at the same time with a single vBackup implementation.

Also, the version of VDDK that you use is dependent on the solution using it (vBackup in this case) rather than the version of ESXi you are running. The current version of vBackup was built around functionality available in VDDK 5.1.4 and 5.5.4 so it is not utilizing any newer functionality that would be available in VDDK 6.0 (if it worked).

Hope this helps.
